genotype variation in h. pylori isolates from iranian patients by rapd-pcr
Authors
abstract
introduction and aims: do h. pylori isolates from normal and dyspeptic patients have similar genetic profiles? since genotype variation occurs within h. pylori population with high frequency, it is tempting to exploit techniques such as rapd-pcr to examine the possible correlation between specific h. pylori genotypes and different peptic diseases. in this study, h. pylori isolates from different dyspeptic patients were genotyped by rapd-pcr. materials and methods: h. pylori isolates from 66 patients, 41 normal, 21 with ulcer, and 4 with cancer were cultured, dnas were extracted by phenol-chloroform. rapd-pcr was optimized, using 10-nt primers of arbitrary sequenceâ (1281, 1254, 1247) and isolate-specific fingerprints were generated. analysis of pcr products on agarose gel was performed using ntsyspc program. dendrograms were calculated according to jaccard & nei. results: according to differences in genetic profiles, h. pylori isolates were clustered into 4 distinct groups: 2 groups consisted of isolates from normal patients, 2 groups of isolates from patients with ulcer, and isolates from patients with cancer were clustered along with isolates from normal a d ulcer patients. furthermore, isolates from ulcer patients appeared in the cluster related to isolates from normal patients. conclusions: genetic variation is quite frequent within h. pylori populations; thus rapd-pcr is an effective technique to reveal genetic diversity of isolates from different dyspeptic patients. in this study, h. pylori strains were clustered into 4 groups: 2 groups from normal patients, and 2 from patients with ulcer. further studies in larger populations will help to correlate a certain peptic disease to specific strain of h. pylori.

Genetic variation among Escherichia coli isolates from human and calves by using RAPD PCR
Background: Various strains of Escherichia coli (E. coli) are known as major causes of intestinal and extraintestinal infections in humans and various animal species. Molecular methods are important for the identification of bacterial isolates and nucleotide sequence variations, as well as information on tracking bacterial agents related to the outbreaks, the frequency of the bacterial genetic ...
full textdna fingerprinting of h. pylori isolates from iranian patients by rep-pcr
introduction and aims: h. pylori has been implicated in peptic diseases, some with detrimental consequences such as ulcer or cancer. since considerable genetic heterogeneity has been observed within h. pylori population worldwide, it appears an ideal achievement to recruit pcr-based methods and design genetic markers which recognize isolates from normal and symptomatic individuals. in this stud...
full textDNA diversity among clinical isolates of Helicobacter pylori detected by PCR-based RAPD fingerprinting.
The RAPD (or AP-PCR) DNA fingerprinting method was used to distinguish among clinical isolates of Helicobacter pylori, a bacterium whose long term carriage is associated with gastritis, peptic ulcers and gastric carcinomas. This method uses arbitrarily chosen oligonucleotides to prime DNA synthesis from genomic sites to which they are fortuitously matched, or almost matched. Most 10-nt primers ...
full textGenetic Profiling of Pseudomonas aeruginosa Isolates from Iranian Patients with Cystic Fibrosis Using RAPD-PCR and PFGE
Objective(s) Pseudomonas aeruginosa is the most important cause of chronic lung infections and death in patients with cystic fibrosis. Determining the distribution of specific strains within patient populations is important in order to examine the epidemiology of the disease and the possibility of cross infection among patients. Materials and Methods Forty six Iranian patients with cystic fib...
full textAnalysis of PCR-RAPD DNA and antibiotic susceptibility profiles of antrum and corpus isolates of Helicobacter pylori from Malaysian patients.
This study was conducted to determine whether there was any genetic heterogeneity among Helicobacter pylori strains isolated from the antrum and corpus of the same individual in a Malaysian population and to determine the presence of heterogeneous susceptibility of the isolates by comparing PCR-RAPD and antibiotic profiles. Forty-four H. pylori isolates cultured from the antrum and corpus of 22...
full textDiscrimination of Dominant H. pylori Strains Isolated from Patients with Different Gastroduodenal Pathologies by Protein Profiling
It is not clear what factors determine divergent outcomes of infections caused by H. pylori . In the present study, the protein profiles of different strains of H. pylori, isolated from three groups of patients with ulcerative disease, non-ulcerative gastritis and cancer disease, were analyzed using 1DSDS-PAGE. The patterns of different H. pylori strains were highly divergent. About 30.76% (7 b...
